Under ISO 11737-1, a simple microbial count is not enough; you must prove that your extraction method actually removes the microorganisms from the device. Bioburden Validation (also known as Recovery Efficiency Testing) is the process of determining how effective a specific laboratory method is at „finding” the microbes on your product. Why Bioburden Validation is Critical
The primary aim of this study is to determine the bioburden recovery factor for your specific test article. Without this validation, your reported bioburden might be significantly underestimated, which can lead to:
- Sterilization Failures: If the recovery factor is low, you may be under-calculating the microbial challenge for your sterilization process.
- Regulatory Non-Compliance: Notified Bodies and the FDA require validated recovery methods to accept bioburden reports.
- Incorrect Safety Margins: Accurate data is essential for establishing appropriate Sterility Assurance Levels (SAL).
Methodology: Determining the Recovery Factor
The bioburden recovery factor is assessed through a controlled inoculation and extraction process:
- Inoculation: A known quantity of specific microorganisms is applied (inoculated) directly onto the sterile test item.
- Extraction: The microorganisms are then removed from the test item using the same extraction methods intended for routine testing (e.g., rinsing or ultrasonication).
- Culturing & Enumeration: The removed microorganisms are cultured and counted to see how many were successfully recovered.
- Calculation: We compare the number of recovered microbes to the number originally inoculated to establish the Recovery Factor (Correction Factor).
Sample Requirements & Timeline
To perform an accurate validation, we require sterile samples to ensure that no pre-existing microbes interfere with the inoculated test strain.
- Minimum Quantity: At least 3 sterile samples are required.
- Turnaround Time: The duration is determined upon the specific study parameters and incubation requirements.
- Device Condition: Samples must be provided in their final, sterile state.
